Experimental drug daraxonrasib shows major benefit in pancreatic cancer trial
NEJM-published phase 1, 2 results show daraxonrasib, a first-in-class RAS(ON) inhibitor from Revolution Medicines, produced meaningful tumor shrinkage and extended survival in patients with advanced RAS‑mutant pancreatic cancer. The trial reported an objective response rate up to 35% and treatment-related grade 3 or higher adverse events in 30% of previously treated patients. Dana‑Farber’s Dr. Brian Wolpin was a lead investigator on the study, and Revolution Medicines has secured FDA clearance for an expanded access program as it advances toward randomized phase 3 testing. Because pancreatic cancer has had few effective targeted options and RAS mutations drive most cases, daraxonrasib could change the standard of care if phase 3 trials confirm these results.
